The assay has been AOAC-RI and AFNOR (ISO 16140-2): validated on a range of food, including dairy samples as well as low and high sample sizes of raw meat (excluding poultry), vegetables, fruits and juices.It enables users to reliably detect and differentiate Shiga-toxin producing Escherichia coli O157:H7 and other STEC in as little as 10 hours.

The Thermo Scientific™ SureTect™ Escherichia coli O157:H7 and STEC Screening PCR Assay is part of a complete workflow for the rapid detection of Escherichia coli O157:H7, stx, eae, and the Escherichia coli STEC serotypes (O26, O45, O103, O111, O121, O145) from the tested samples. 

Samples assessed as positive using the SureTect™ Escherichia coli O157:H7 and STEC Screening PCR Assay are further tested using this SureTect™ Escherichia coli STEC Identification PCR Assay.

Thermo Scientific™ SureTect™ Escherichia coli O157:H7 and STEC Screening PCR Assay includes:

• SureTect Lysis Reagent 1 Tubes
• SureTect Proteinase K
• SureTect Escherichia coli O157:H7 and STEC Screening PCR Tubes
• ThermoScientific 96 well pierceable seals
• SureTect PCR Caps

Enriched samples are pipetted into pre-filled lysis tubes, along with Proteinase K, before incubation, to lyse any bacterial cells present in the sample and release their DNA into the solution. The lysate is then loaded into the SureTect Escherichia coli O157:H7 and STEC Screening PCR tube to re-hydrate the pre-loaded PCR pellet which contains all the necessary components and reagents for PCR, including a probe, primers and DNA template for the internal amplification control (IAC).

The PCR tubes are then sealed, loaded into Applied Biosystems™ QuantStudio™ 5 Food Safety Real Time PCR instrument used with RapidFinder™ Analysis Software v2.0or later. After completing the run, interpreted results are clearly displayed and can be reported, stored, printed or downloaded as required. The internal amplification control confirms correct assay conditions have occurred.
