Description

For use in NucleoSpin™ Plasmid DNA purification kits for rapid small-scale preparation of high-copy plasmids from E. coli. Macherey-Nagel™ NucleoSpin™ Resuspension Buffer A1 without RNase A is a replacement component used in the NucleoSpin™ method's cell lysis process to resuspend the cell pellet.

The NucleoSpin™ Plasmid kit is designed for purifying up to 20μg of high copy-number plasmid DNA from 1mL to 5mL E. coli overnight cultures preferably grown in LB medium. The NucleoSpin™ Plasmid binding strips contain a unique silica membrane that binds DNA in the presence of chaotropic salt.

After a modified alkaline lysis procedure, bacterial lysates are loaded onto the NucleoSpin™ Plasmid filter strips arranged on the NucleoVac 96 vacuum manifold. Filtrates are collected directly in the NucleoSpin™ Plasmid binding strips. After binding and washing, highly pure plasmid DNA is eluted in low-salt buffer (5mM Tris/HCl, pH 8.5). Alternatively, water (pH 8.5) can be used.

Kit components: NucleoSpin™ Plasmid filter strips, NucleoSpin™ Plasmid binding strips, all necessary buffers, RNase A, MN wash plate, MN tube strips, cap strips.
