Beschreibung
- The assay recognizes mouse TNFSF11. An antibody specific for TNFSF11 has been pre-coated onto the microwells. The TNFSF11 protein in samples is captured by the coated antibody after incubation.
- Following extensive washing, another antibody of biotinylated specific for TNFSF11 is added to detect the captured TNFSF11 protein.
- For signal development, Streptavidin-HRP is added, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm
- TNFSF11 also known as RANKL, is a member of the tumor necrosis factor (TNF) cytokine family which is a ligand for osteoprotegerin and functions as a key factor for osteoclast differentiation and activation.
- RANKL is produced by a diverse set of lineage-specific cells and is involved in a wide variety of physiological processes that include skeletal remodeling, lymph node organogenesis, mammary gland development, and thermal regulation.
- RANKL was shown to be a dendritic cell survival factor and is involved in the regulation of T cell-dependent immune response.
- The deficient mice exhibited defects in early differentiation of T and B lymphocytes, and failed to form lobulo-alveolar mammary structures during pregnancy.
