Solid phase sandwich ELISA used to detect and quantify protein levels of endogenous SELP

  • Recognizes human SELP
  • SELP (P-Selectin, GMP-140, CD62P) is a transmembrane glycoprotein that mediates interaction of activated endothelial cells or platelets with leukocytes
    • Adhesion molecule involved in pathogenesis of inflammation, thrombosis, oncogenesis
    • Stored in alpha-granules of platelets and Weibel-Palade bodies of endothelial cells
    • Upon cell activation by agonists, P-selectin is transported to cell surface

Process:

  • Antibody specific for SELP pre-coated onto microwells
  • SELP protein in samples captured by the coated antibody after incubation
  • After extensive washing, another antibody specific for SELP is added to detect captured SELP protein
  • For signal development, horseradish peroxidase (HRP)-conjugated antibody is added, followed by Tetramethyl-benzidine (TMB) reagent
  • Solution containing sulfuric acid used to stop color development and color intensity
    • Proportional to quantity of bound protein, measurable at 450 nm with correction wavelength set at 630 nm
