Description

Solid phase sandwich ELISA used to detect and quantify protein levels of endogenous KIM-1

  • Recognizes human KIM-1
  • KIM-1 (Kidney injury molecule 1, Hepatitis A virus cellular receptor 1, HAVCR, T-cell immunoglobulin, mucin domain 1, TIM-1) is a class I integral membrane glycoprotein with ectodomain containing Ig-like domain and mucin domain
    • Acts as membrane receptor for HAV
    • Provides costimulatory signal for T cell activation and inhibits development of peripheral tolerance
    • Involved in regulation of asthma and allergic diseases
    • Reported KIM-1 shed into urine after acute kidney damage
    • Marker of renal tubular injury

Process:

  • Antibody specific for KIM-1 pre-coated onto microwells
  • KIM-1 protein in samples captured by the coated antibody after incubation
  • After extensive washing, another antibody specific for KIM-1 is added to detect captured KIM-1 protein
  • For signal development, Streptavidin-HRP is added, followed by Tetramethyl-benzidine (TMB) reagent
  • Solution containing sulfuric acid used to stop color development and color intensity
    • Proportional to quantity of bound protein, measurable at 450 nm with correction wavelength set at 630 nm
