Description

Solid phase sandwich ELISA used to detect and quantify protein levels of endogenous IL15

  • Recognizes human IL15
  • IL15 (Interleukin 15) is a cytokine important for development, maturation and function of many cells of immune system
    • Expression affects homeostasis and function of B cells through NK cell-derived IFNG
    • High levels of expression associated with pathogenesis of autoimmune and inflammatory diseases (Chron's disease, psoriasis, leukemias, RA, graft rejection)

Process:

  • Antibody specific for IL15 pre-coated onto microwells
  • IL15 protein in samples captured by the coated antibody after incubation
  • After extensive washing, another antibody of biotinylated specific for IL15 is added to detect captured IL15 protein
  • For signal development, Streptavidin-HRP is added, followed by Tetramethyl-benzidine (TMB) reagent
  • Solution containing sulfuric acid used to stop color development and color intensity
    • Proportional to quantity of bound protein, measurable at 450 nm with correction wavelength set at 630 nm
