Description

Solid phase sandwich ELISA used to detect and quantify protein levels of endogenous IDH1

  • Recognizes human IDH1
  • IDH1 (PICD, IDP) belongs to isocitrate and isopropylmalate dehydrogenases family
    • IDH1 may be tumor suppressor, when mutationally inactivated, contributes to tumorigenesis in part through induction HIF-1 pathway
    • Common feature of major subset of primary human brain cancers
    • IDH1 mutation heterozygotic, functions as dimer

Process:

  • Antibody specific for IDH1 pre-coated onto microwells
  • IDH1 protein in samples captured by the coated antibody after incubation
  • After extensive washing, another antibody specific for IDH1 is added to detect captured IDH1 protein
  • For signal development, horseradish peroxidase (HRP)-conjugated antibody is added, followed by Tetramethyl-benzidine (TMB) reagent
  • Solution containing sulfuric acid used to stop color development and color intensity
    • Proportional to quantity of bound protein, measurable at 450 nm with correction wavelength set at 630 nm
