Solid phase sandwich ELISA used to detect and quantify protein levels of endogenous CXCL10

  • Recognizes human CXCL10
  • CXCL10 (IP-10) is a member of CXC chemokine family that binds to CXCR3 receptor to exert its biological effects
    • 12-kDa protein, constitutes and constitutes two internal disulfide cross bridges
    • Cleavage generates a 10-kDa secreted polypeptide with four conserved cysteine residues in N-terminal
    • CXCL10 mediates lekocyte trafficking, adaptive immunity, inflammation, haematopoiesis and angiogenesis

Process:

  • Antibody specific for CXCL10 pre-coated onto microwells
  • CXCL10 protein in samples captured by the coated antibody after incubation
  • After extensive washing, another antibody of biotinylated specific for CXCL20 is added to detect captured CXCL10 protein
  • For signal development, Streptavidin-HRP is added, followed by Tetramethyl-benzidine (TMB) reagent
  • Solution containing sulfuric acid used to stop color development and color intensity
    • Proportional to quantity of bound protein, measurable at 450 nm with correction wavelength set at 630 nm
