Solid phase sandwich ELISA used to detect and quantify protein levels of endogenous B7-H3/CD276

  • Recognizes human B7-H3/CD276
  • B7-H3 (CD276) is a Type I transmembrane protein expressed on many tissues and cell types
    • B7-H3 is a 100-kDa glycoprotein belonging to B7 immunoregulatory family
      • Participates in regulation of T-cell-mediated immune response by functioning as T cell costimulator and coinhibitor
    • Overexpressed on human solid cancers, implicated in cancer progression and metastasis, target for cancer immunotherapy

Process:

  • Antibody specific for B7-H3/CD276 pre-coated onto microwells
  • B7-H3/CD276 protein in samples captured by the coated antibody after incubation
  • After extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for B7-H3/CD276 is added to detect captured B7-H3/CD276 protein
  • For signal development, followed by Tetramethyl-benzidine (TMB) reagent
  • Solution containing sulfuric acid used to stop color development and color intensity
    • Proportional to quantity of bound protein, measurable at 450 nm with correction wavelength set at 630 nm
