Description

Solid phase sandwich ELISA used to detect and quantify protein levels of endogenous APCS

  • Recognizes human APCS
  • Serum amyloid P component (SAP, APCS) belongs to family of pentraxins, a superfamily of plasma proteins characterizes by pentameric assembly and calcium-dependent ligand binding
    • Constitutive serum protein synthesizes by hepatocytes
    • Can interact with DNA and histones, may scavenge nuclear material released from damaged circulating cells
    • Can bind to fibrils in all types of amyloid deposits and contributes to pathogenesis of amyloidosis

Process:

  • Antibody specific for APCS pre-coated onto microwells
  • APCS protein in samples captured by the coated antibody after incubation
  • After extensive washing, another antibody specific for APCS is added to detect captured APCS protein
  • For signal development, Streptavidin-HRP is added, followed by Tetramethyl-benzidine (TMB) reagent
  • Solution containing sulfuric acid used to stop color development and color intensity
    • Proportional to quantity of bound protein, measurable at 450 nm with correction wavelength set at 630 nm
