Quantitative determination of human IL-6 in serum, plasma, and cell culture supernatant

IL-6 specific monoclonal antibody binds to immobilized antibody coated on 96-wells plate. Wells are washed and streptavidinhorseradish peroxidase conjugate mixed with biotinylated anti-human IL-6 antibody is added, producing an antibody-antigen-antibody “sandwich”. The wells are again washed and TMB substrate solution is added, which produces a blue color in direct proportion to the amount of IL-6 present in the initial sample. The Stop Solution changes the color from blue to yellow, and the microwell absorbances are read at 450nm.
