For quantitative determination of human IL-4 in serum, plasma, and cell culture supernatant

Utilizes monoclonal antibody specific for IL-4 coated on 96-well plate that binds to immobilized antibody. Wells are washed and Streptavidin-horseradish peroxidase conjugate mixed with biotinylated anti-human IL-4 antibody is added, producing antibody-antigen-antibody “sandwich”. Wells are again washed and TMB substrate solution is added, which produces blue color in direct proportion to amount of IL-4 present in initial sample. The stop solution changes the color from blue to yellow, and microwell absorbances are read at 450nm
