Microplate reader capable of measuring absorbance at 450nm, precision pipettes to deliver 50μL and 100μL volume, adjustable 1mL, 5mL, 10mL, 25mL pipettes for reagent preparation, deionized or distilled water, wash bottle or automated microplate washer, log-log graph paper or automated data reduction, tubes to prepare standard dilutions, laboratory timer, absorbent paper
Utilizes monoclonal antibody specific for human C4a/C4a-desArg coated on 96-well plate. Standards and samples are added to wells, and any C4a-desArg present binds to immobilized antibody. The wells are washed and a mixture of biotinylated polyclonal anti-human C4a antibody and streptavidin-horseradish peroxidase is added, producing an antibody-antigen-antibody “sandwich”. The wells are again washed and substrate solution is added, which produces a blue color in direct proportion to the amount of C4a-desArg present in initial sample. Stop Solution changes the color from blue to yellow, wells are read at 450nm
