Are You Looking to Improve Sensitivity for Rare DNA and RNA Targets During Your qPCR Testing?

New technologies like the Applied Biosystems™ QuantStudio™ Absolute Q™ dPCR System and MAP16 Microfluidic Plates bring innovation to the lab and allow scientists pooling different sample data to detect very low abundance targets, increasing sensibility and limit of detection. While PCR sample input volume can affect your ability to detect ultra-rare targets, even those with a mutant allele frequency (MAF) of 0.1 percent can be detected using multiple dPCR arrays on a QuantStudio™ MAP16 plate using the digital pooling feature of the Absolute Q™ Analysis Software.

  • Digital pooling, which combines data from multiple dPCR arrays to enable analysis of a grerater sample volume than conventional quantitative PCR

The analysis of a MAF DNA mixture (0.1 percent, 27μL) using four combined arrays on a QuantStudio™ MAP16 plate (36μL of dPCR mix) could be used for a multitude of rare-target detection applications in the precision medicine space, such as for monitoring treatment response, screening for minimal residual disease, and rapid identification of mutation-linked drug resistance.

MAP16 Microfluidic Plates are a key component of the QuantStudio™ Absolute Q™ dPCR System and are powered by patented microfluidic array plate (MAP) technology.

Highlights include:

  • Consistent, automated separation of a sample into 20,000 microchambers with an industry-leading, less than 1 percent coefficient of variation (CV)
  • Microinjection-molded plates to overcome inconsistency and high dead volume, allowing precise microchamber volume and consistent sample distribution, and thus analysis of greater than 98 percent of loaded volume
  • Fixed microchamber arrays and positive pneumatic pressure to distribute (or digitize) reagents for dPCR, as opposed to other methods like digital droplet PCR that use fluid shearing to generate droplets that are less homogeneous and more variable in delivering results
  • Plates consisting of a 4 x 4 grid of dPCR arrays, each of which contains 20,480 fixed microchambers. A design that allows flexibility in the number of samples or arrays run simultaneously without sacrificing data quality
