Description
- Recognizes human HSP90 protein
- HSP90 encoded by HSP90AA1 is a constituitively and ubiquitously expressed molecular chaperone is crucial for stability and function of proteins
- Provides chaperoning activity for client proteins, implication in tumor malignancy
- Mainly residues in cytosol, can be released into extracellular space
- Level of plasma HSP90 is positively correlated with tumor malignancy in clinical cancer patients, promising diagnostic marker for tumor malignancy in clinical application
Process:
- Antibody specific for HSP90 pre-coated onto microwells
- HSP90 protein in samples captured by the coated antibody after incubation
- After extensive washing, another antibody specific for HSP90 is added to detect captured HSP90 protein
- For signal development, horseradish peroxidase (HRP)-conjugated antibody is added, followed by Tetramethyl-benzidine (TMB) reagent
- Solution containing sulfuric acid used to stop color development and color intensity
- Proportional to quantity of bound protein, measurable at 450 nm with correction wavelength set at 630 nm
