Description

The NucleoSpin™ 96 Flash kit provides rapid, high-throughput isolation of high copy plasmid, cosmid and BAC DNA using vacuum manifolds or centrifuges and ensures reproducible yields of DNA in the convenient 96 well format. Ideal for high-throughput automated sequencing projects. With the NucleoSpin™ 96 Flash method plasmid is liberated from theE. colihost cells by SDS/alkaline lysis after resuspension of the bacterial pellets. The resulting lysate is then neutralised and cell debris precipitates together with SDS. Optional incubation in boiling water inactivates nucleases that are present in a variety ofE. colihost strains and denatures soluble proteins and other cell constituents. All precipitates are then removed by filtration of the lysate through the NucleoSpin™ Flash filter plate. Plasmid DNA is collected by addition of isopropanol to the filtrate and subsequent centrifugation. After an additional washing step with 70% ethanol and drying the DNA can be resuspended in 5mM Tris-Cl, pH8.5.Kit components: NucleoSpin™ Flash filter plates, square well blocks and cover sheets for bacterial cultivation, square well blocks for precipitation, adhesive aluminum foils, buffers, RNase A.
